BACKGROUND: The proteomic profile of cryopreserved in vitro produced bovine embryos is little known but can provide insights on the successful application of cryo procedures in support of animal breeding. OBJECTIVE: To identify embryonic proteins and biomarkers related to improved cryotolerance of vitrified in vitro produced bovine embryos. MATERIALS AND METHODS: Proteins were isolated from embryo pools (n = 25 embryos per replicate) and analyzed using the nanoLC - MS/MS system. Further, the UniProtKB database (Uniprot -http://www.uniprot.org/) was used for protein identification. Proteins were classified based on their molecular mass, isoelectric point, and enzymatic activity. Post-translational modification predictions and functional gene ontology analysis were performed as well. Finally, a protein-protein interaction network was created to shed light on the embryo interactome. RESULTS: Based on the MS/MS approach, 66 proteins were identified from vitrified Bos taurus embryos. The retrieved proteins were presumably annotated, which allowed a description of the qualitative and functional aspects of the embryo proteome after the vitrification process. CONCLUSION: These findings allowed us to conclude that in vitro-produced vitrified embryos expressed proteins that underlie biological processes related to reproduction, stress and lipid metabolic process, which are essential to maintain embryo viability. doi.org/10.54680/fr22410110512.

The predatory stink bugs are well known by their behavior, but the knowledge of the immature morphology and their natural history are scarce. Studies on predatory stink bugs are important to better understand their evolution and their use as biological controllers. Here, we describe the morphology of egg and the five nymphal instars of Oplomus catena (Drury, 1782), using optical and scanning electron microscopy. In general, O. catena immatures are very distinctive from other Asopinae species already studied. The egg is black, with short aero-micropylar processes and similar to those described for Stiretrus species. The nymphs can be diagnosed by the abdominal plates very large and bright blue. The color polymorphism of adults is fully illustrated, and four color patterns are proposed. The natural history of the species is described based on field and laboratory observations. The known prey of the species is reviewed and new preys are reported. The morphological and biological traits here described are discussed in order to better understand the biological role of predatory stink bugs.

The aim of this study was to evaluate the supplementation of embryo culture medium with antioxidant obtained from oily extract of Lippia origanoides on in vitro blastocyst development and quality. Oocytes collected from slaughterhouse ovaries were matured and fertilized in vitro following standard laboratory procedures. Zygotes were cultured in SOF medium supplemented according to the following treatments: T1 embryo culture medium without antioxidant supplementation; T2)50µM/mL Cysteamine; T3)2.5µg/mL; T4)5.0µg/mL and T5)10.0µg/mL of antioxidant obtained from oily extract of Lippia origanoides. On the seventh day of culture, the blastocysts were fixed and evaluated for apoptosis rates, number of total cell and inner cell mass cells by means of the TUNEL Test. The use of antioxidants during cultivation did not increase (P> 0.05) the final blastocyst production rate. The treatments T2, T3, T4 and T5 had the lowest (P< 0.05) apoptotic indexes (4.5±1.1%, 8.4±2.5%, 3.4±1.1% and 5.5±0.9%, respectively) when compared to T1 treatment (10.0±1.4%). The number of inner cell mass did not differ (P> 0.05) among embryos from different treatments. The addition of antioxidant obtained from oily extract of Lippia origanoides reduces the apoptosis rate and improves the quality without increasing the total in vitro production of bovine embryos.(AU)


O objetivo desse estudo foi avaliar a suplementação de meio de cultura de embriões com antioxidante obtido do extrato oleoso da Lippia origanoides no desenvolvimento e na qualidade de blastocistos produzidos in vitro. Oócitos coletados de ovários de matadouros foram maturados e fertilizados in vitro segundo procedimento laboratorial padrão. Zigotos foram cultivados em meio SOF suplementado de acordo com os seguintes tratamentos: T1) meio de cultivo embrionário sem suplementação antioxidantes; T2) 50µM/mL Cisteamina; T3) 2,5µg/mL; T4) 5,0µg/mL e T5) 10,0µg/mL do antioxidante obtido do extrato oleoso de Lippia origanoides. No sétimo dia de cultivo, os blastocistos foram fixados e avaliados para taxa de apoptose, número total de células e massa celular interna através do teste TUNEL. O uso de antioxidantes durante cultivo não aumentou (P>0,05) a taxa de produção final de blastócitos. Os tratamentos T2, T3, T4 e T5 tiverem menor índice apoptótico (p>0,05 - 4,5±1,1%, 8,4±2,5%, 3,4±1,1% e 5,5±0,9%, respectivamente) quando comparados a T2 (10,0±1,4%). O valor de massa celular interna não diferenciou (p>0,05) entre embriões de diferentes tratamentos. A adição de antioxidante obtido do extrato oleoso de Lippia origanoides reduziu a taxa de apoptose e melhorou a qualidade sem aumentar a produção in vitro de embriões bovinos.(AU)

Canola (Brassicaceae: Brassica spp.) is an important feedstock for biodiesel production and a potential ingredient for use in the food industry. In different continents, various arthropod pests damage canola plants. Stink bugs (Heteroptera: Pentatomidae) are present in all zoogeographical regions, and many species are recognized by their economic importance as crop pests. Our aim was to describe the composition, structure, and diversity of the assemblage of stink bugs sampled on canola in southern Brazil. A total of 878 pentatomids were captured, belonging to 27 species. The dominant species were Euschistus heros (F.) (n = 439), Dichelops furcatus (F.) (n = 160), and Nezara viridula L. (n = 79). The species richness estimators indicated the samples correspond from 79.7 to 93.1% of the richness estimated. Comparing canola to other monocultures, the richness of Pentatomidae was much superior. Otherwise, when compared to studies conducted in native vegetation and urban fragments, the richness is similar, and sometimes higher. This relatively high number of associations brings out the vulnerability of the canola fields according to the expansion of its cultivated area. Nine species of Pentatomidae are recorded on canola for the first time in Brazil. The most abundant species reported here are often stressed as stink bugs of economic importance in agro-ecosystems. The economic importance of each species varies greatly depending on the plant attacked. Detailed studies are needed to evaluate the damage caused to canola by stink bugs.

The aim of this study was to evaluate the pattern of expression of LHCGR isoforms in Gir heifers characterized as good (10.3 ±â€¯1.2 ova/embryos per flush, n = 5) or poor responders (1.1 ±â€¯0.3 ova/embryos per flush, n = 5) to superovulation protocols. In both groups, an adapted ultrasound-guided follicular aspiration system was used to collect granulosa cells from 8 mm follicles formed either during a synchronized, non-stimulated follicular wave (no stimulation control, NS) or on the fourth day of a superovulation protocol (SOV) induced with 200 IU of pFSH. The recovered follicular fluid was centrifuged and granulosa cells were washed with NaCl 0.9% and kept in RNAlater®. RNA extraction was performed using a commercial RNeasy Micro Kit and eluted samples were quantified and reverse transcribed using the commercial Superscript III kit. cDNA samples were amplified by real-time PCR using a primer to target LH/hCG receptor gene - not selective for LHCGR isoforms (total LHCGR) - and four sets of isoforms selective primers (S1, S10, S10 + 11, and S11). Analyses were performed using the REST software and expression levels are shown as mean ± SEM. Under physiological conditions (NS), poor responders had a higher expression of total LHCGR (4.9 ± 1.7 fold-change, P < 0.01) as well as isoforms S10, S11 and S10 + 11, compared to good responders. In both phenotypes, superovulation down-regulated total LHCGR expression (-0.5 ± 0.2 and -0.9 ± 0.0 for good and poor responders, respectively; P < 0.05). However, in poor responders the exogenous FSH treatment up-regulated the S10 (2.4 ± 2.0; P < 0.05), S10 + 11 (3.8 ± 3.2; P < 0.01), and S1 isoforms (1.8 ± 1.3; P < 0.05), compared to good responders We conclude that down-regulation of total LHCGR, associated to up-regulation of their inactive isoforms, may have compromised follicle development and thus contributed to the low efficiency of superovulation in heifers with a poor responder phenotype.

In broiler breeder roosters, the weight of the testes is positively associated with daily sperm production and fertility. In birds, the testes are located in the coelomic cavity, not being accessible to reproductive evaluation as mammalian testes. The reproductive evaluation of roosters is based on phenotypic traits. Any changes on testes will be reflected on fertility levels. The aim of this paper was to evaluate the potential of association of phenotypic traits as body weight (BW), comb scores (COS), cloaca scores (CLS) and feet scores (FS) with testicular morphometric parameters in adult Cobb® Mx roosters in two ages (25 and 45 weeks of age). The low BW roosters had lower testes weight (TW), smaller seminiferous tubule diameter (STD) and lower comb and cloaca scores than the medium and high BW roosters (P≤0.05). Heavy roosters had the highest COS and bigger seminiferous epithelium height (SEH). There was no statistical difference between the BW categories in FS and volumetric proportion of seminiferous tubule (ST) and interstitial tissue (IT). Considering the age effect, roosters with 45 weeks of age had lower TW and ST than 25-week roosters. Positive, moderate and significant correlation was found between testes weight and phenotypic characteristics as BW, COS and CLS in 45week roosters. A positive, moderate and significant correlation was found between body weight and histologic testicular characteristics as STD and SHE in both evaluated ages. In conclusion, there is a regression in the testes weight with age and it is more intense in Cobb® Mx roosters with lower BW. Furthermore, it is concluded that the BW positively influences the testes weight and histological quality of the testes. COS and CLS can be used with moderate potential prediction to identify roosters with low testes weight at 45 weeks of age.(AU)


Em galos de matriz pesada sabe-se que o peso dos testículos pode ser associado positivamente com a produção diária de espermatozoides e que alterações nos testículos irão refletir na fertilidade. Nas aves, os testículos estão localizados dentro da cavidade celomática, e, portanto, não são acessíveis para exame andrológico direto, como em mamíferos. A avaliação reprodutiva de galos se baseia principalmente em características fenotípicas. Buscou-se avaliar o potencial de associação de alguns parâmetros fenotípicos, como peso corporal (PC), escores de crista (ECR), escores de cloaca (ECL) e escores de pés (EP), com parâmetros morfológicos de testículos em galos Cobb® Mx em duas idades (25 e 45 semanas de idade). Galos leves apresentaram menor peso testicular (PT), menor diâmetro de túbulo seminífero (DT), e menores ECR e ECL que galos médios e pesados (P≤0.05). Galos pesados apresentaram os maiores ECR e maiores valores para altura do epitélio seminífero (AE), e DT. Não houve diferença estatisticamente significativa entre as categorias de peso para escores de pé e proporções de túbulo seminífero (TS) e interstício (INT). Considerando-se o efeito da idade, galos com 45 semanas de idade apresentaram menores PT e TS que galos com 25 semanas de idade. Correlação positiva, moderada e significativa foi encontrada entre peso de testículo e características fenotípicas como ECR, ECL e PC em galos de 45 semanas de idade. Foi encontrada correlação positiva, moderada e significativa entre PC e características testiculares como DT e AE nas duas idades avaliadas. Conclui-se que em galos Cobb® Mx ocorre regressão do testículo com o avanço da idade, e ela é mais intensa em galos com menor peso corporal. Conclui-se que o PC tem influência positiva no peso e índices histomorfométricos dos testículos, e que escores de crista e de cloaca podem ser utilizados com potencial moderado de predição para identificar galos com baixo peso de testículos em galos com 45 semanas de idade.(AU)

O Brasil atualmente é detentor do terceiro maior rebanho de vacas leiteiras do mundo, composto, em sua maioria, de animais mestiços F1 (Gir X Holandês), os quais são bem adaptados às áreas tropicais. O objetivo do presente estudo foi investigar a eficiência da ovum pick-up (OPU) e da produção in vitro de embriões oriundos de doadoras Girolando com sêmen sexado de touros provenientes de duas raças, Gir e Holandesa. Foram utilizados dados referentes a 232 animais de diversos rebanhos, totalizando 4334 oócitos recuperados. Os oócitos foram classificados, e as estruturas viáveis (GI, GII e GIII) foram utilizadas para produção in vitro de embriões (PIVE). Foi observada uma média de oócitos recuperados para as doadoras F1 de 18,14±1,33. Além disso, notou-se que não houve diferença entre os dois cruzamentos utilizados, considerando média de total de embriões (4,96±0,40 e 6,56±0,76) para o cruzamento F1 X HPB (Holandês preto e branco) e F1 X Gir, respectivamente. Portanto, as doadoras F1 apresentaram potencial como doadoras de oócitos no sistema de produção in vitro de embriões, independentemente da raça do touro cujo sêmen foi utilizado.(AU)


Brazil currently has the third largest herd of dairy cattle in the world, composed mainly by F1 crossbreed animals (Gyr x Holstein), which are well adapted to tropical areas. The aim of this study was to evaluate the efficiency of Girolando donors in OPU and in vitro production of embryos (IVEP) using sexed semen of bulls from two breeds, Gyr and Holstein. We used data from 232 animals coming from different herds and the total of recovered oocytes was 4334. The oocytes were classified according to their viability and the viable ones (GI, GII and GIII) were used for IVP. The average of oocyte recovery from F1 donors was 18.14±1.33. There was no difference on the average of embryos considering the crossing F1 X Holstein (4.96±0.40) and F1 X Gyr (6.56±0.76). In conclusion, F1 donors showed a potential for being oocyte donors in the system of in vitro production of embryos, regardless the breed of the bull.(AU)

Heteroptera is the most numerous and diverse suborder of Hemiptera, with about 38,000 species. This diversity also involves cytogenetic features, including chromosome number and a sex determining system. Information about heterochromatin occurrence and distribution is scarce in heteropterans, but still, there is some evidence of variability. We determined the chromosome number and CMA3/DAPI-banding pattern of 179 individuals of 25 heteropteran species from Brazil. Eight species of Pentatomidae exhibited a constant chromosome number (2n = 12 + XY), but in Coreidae (12 species), Largidae (1 species), Rhopalidae (1 species), and Pyrrhocoridae (3 species), the numbers ranged from 2n = 10 + 2m + X0 to 2n = 24 + 2m + X0. Although there were no large differences in the chromosome size between species, the CMA3/DAPI-banding patterns differed markedly. Among the genera, species of Edessa, Spartocera, Hypselonotus, Phtia,Holhymenia and Euryophthalmus showed a large accumulation of heterochromatin, while the other species exhibited few or no heterochromatic bands. In general, when heterochromatin was more accumulated, this occurred preferentially at terminal positions, except in Holhymenia histrio, which exhibited intercalary bands. This study made it possible to identify some chromosome rearrangements and to enhance our knowledge of the evolutionary mechanisms that determine karyotype differentiation in Heteroptera.

Nymphs and adults of the burrower bug Scaptocoris carvalhoi Becker feed on vegetal sap of their host plants through the roots, and little is known on the morphology and biology of its immature stage. Therefore, we aimed to characterize the immatures of S. carvalhoi by describing the egg and the morphology of each instar. Eggs of S. carvalhoi have a smooth chorion surface without visible micropylar processes. The presence of five instars was confirmed by the coefficient of determination (R (2) > 0.95) and by the growth constant (K between 1.2 and 1.6). Nymphs have an elliptical body and fossorial scythe-like forelegs. The tarsi are absent as in adults, and the prototarsal insertion region becomes visible only in the fourth instar. Nymphs from first to fourth instar of S. carvalhoi showed the presence of 1 + 1 trichobothria in urosternites III to VII, close to the anterior margin and inside the spiracles; besides these trichobothria, fifth instars presented 1 + 1 pre-trichobothria in urosternites III to V located posteriorly, almost in the row of spiracles close to the posterior margin of the urosternites. This is the first detailed morphological record of immatures belonging to Scaptocoris.

RESUMEN

Asopines are predators of insects, with several species with potential as biocontrol agents of a number of pests. Metrogaleruca obscura (Degeer), a neotropical species of Galerucini, was introduced in Malaysia, Asia, and Mauritius, Africa, to control the spread of Cordia curassavica (Boraginaceae), a native plant of the neotropics. The occurrence of Podisus nigrispinus (Dallas) preying on M. obscura is recorded, and Cordia verbenacea (Boraginaceae) is mentioned as a host plant for M. obscura. A list of Chrysomelidae attacked by asopines in the neotropical region is also presented.

RESUMEN

Neem (Azadirachta indica) seedlings were found infested by the brown burrower bug, Scaptocoris castanea Perty, in December 2009, in the county of Tupaciguara, Minas Gerais state. Symptoms observed varied from leaf yellowing and stem drying, reduction in root size and number to plant death. This is the first report of S. castanea attacking neem plants.
